Summary A Research Assistant II (RA-II) primarily conducts cancer-focused research projects for the Advanced Cell Engineering and 3D Models Core facility under the supervision of the shared resource facility directors. The appointed individual will assist in the experimental planning, implementation, and acquisition and/or analysis of research data for Core facility users. The appointee will be conducting experimental procedures involving 3D cancer organoid development and growth, CRISPR/Cas genetic engineering, and xenograft implantation on the chick egg chorioallantoic membrane. The RA-II will also assist with training of other laboratory personnel and/or trainees in experimental techniques and lab SOPs. Other responsibilities include user project entry into the iLab billing system to ensure collection of charge-backs and assistance with yearly progress reports. Nature of certain projects may require occasional weekend work. This includes: CAM user project drug treatment studies, organoid maintenance and drug treatment, lab emergencies involving equipment, etc. Job Duties Growth, mantainance, characterization, biobanking, genetic editing and functional assays using tumor organoids derived from PDXs and/or human patient primary tumors. Assistance with CRISPR/Cas genetic engineering projects including: generating lentivirus, cloning, electroporation, clone growth and isolation, genomic PCR, etc. Chicken egg xenograft implantation of primary patient tumors or patient-derived xenografts on the chorioallantoic membrane (CAM) as a fee-for-service for Core facility users. Following projects through completion (i.e. from tumor xenograft implantation to analysis on assigned projects). Acquisition of data including: imaging tumors on CAM and/or organoids, tumor and relevant tissue dissection for processing, tissue DNA isolation and analysis, qPCR assay and analysis, and detailed experimental procedure recording. Compiles user data reports for assigned cancer-focused projects and will send to Core director for review and distribution to Core users. Training in iLab software will be required and employee will be expected to bill for completed Core user projects. Meets with prospective Core users for project planning consultations as time permits. Assists with research and development projects as time permits including both CAM and tumor organoid growth optimization and characterization. Performs daily tasks (i.e. daily monitoring of chicken egg viability, disposal of biohazardous waste, CO2 tank check and changing, autoclaving lab materials, disinfecting tabletop and cabinet incubators, and maintaining adequate temperature & humidity in incubators) to ensure proper workflow of the Core facility. Assists with tracking lab supply inventory and notifies director of any supplies that need to be ordered. Assists with training of new lab personnel and/or trainees rotating through the lab in proper experimental techniques, troubleshooting, and lab SOPs. Assists Core director with compiling data and reports needed for annual progress reports and/or grant applications. Assembles and delivers presentations for the ACE3M Core lab meetings on research projects that have been completed as well as quarterly journal club presentations. Minimum Qualifications Bachelor's degree in a Basic Science or a related field. Three years of relevant experience. Preferred Qualifications Master's degree in basic science or a related field. Proficient in Microsoft Word, Excel, and Powerpoint. Standard laboratory techniques (i.e. microscopy knowledge, molecular biology techniques - PCR, cloning, ELISA, etc. Proficiency in cell culture required (i.e. cell culture, bacterial culture, spheroid/organoid culture, etc.). ImageJ, Adobe, and/or GraphPad Prism. Standard laboratory cell culture proficiency including 3D culture models. Laboratory animal work experience.
